The history of Zentiva began in 1488 with the opening of a small pharmacy in the centre of Prague. The co-owners of the pharmacy then built a modern pharmaceutical factory in Dolní Mecholupy (now part of Prague). In 1930, it began manufacturing medicines, and in 1946, the Czech government nationalised Zentiva. It was merged with the state-owned pharmaceutical company SPOFA, which led to the separation of the pharmacy and drug manufacturing businesses.

From 2003 to 2006, Zentiva acquired three pharmaceutical companies, and in 2008 it became part of Sanofi. By 2020, the company had acquired several industrial production facilities.

Since 2004, the company's products have been sold in France, England, Poland, and since 2007, in Turkey.
 

Today, Zentiva is a leader in the production and sale of branded generics and over-the-counter products in Central and Eastern Europe, Ukraine, and Turkey. Zentiva's drugs for the treatment of heart and vascular diseases, cancer, diabetes, thrombosis, central nervous system diseases, as well as vaccines, are recognised as some of the best in the world.
